In the Xplay video if you have sex with one chick the other ones are going to react accordingly. Judging by the vid where you see Subject 0 fighting with Miranda, if you get it on with Miranda Subject 0 might hate you, and vice versa. We’ll see how it plays out.
I imagine Ashley is only on that planet if you saved her in 1. Kaiden would be there if the other option were chosen.
Regarding romances in this game, any romances pursued here will affect your previous romance when you do run into them. It will also affect them when you get them back into your party in Mass Effect 3.

I’d preload it on steam and get hype monday at 9pm, cause my schedule is loose on tuesday. But I guess I’m gonna be stuck just watchin my roomate play his 360 ver of ME2 if he gets it day 1. I was thinking about taking my incomplete Insanity Adept/Bastion Shep, or using my old Infiltrator Shep that was maxed intimidate and who offed the krogan, and had let the engineer die over the soldier.
